You caught a glimpse of Betancourt's problems (lack of opportunities and his own misdoings) last week. Following his June 20 botch job that put him under the microscope, he ran off three straight scoreless appearances (two saves), although he had to work around a pinch of trouble in the last two.

He slipped up again on Thursday, allowing a leadoff jack to Bryce Harper that tied the game and produced Betancourt's fourth blown save in 16 tries this year, including his third of the month. It was the third tater he allowed in 2012, and he's about on pace to meet last year's seven. In reality, he should be on pace to trump that; aerial vulnerability marks his chief weakness.

His four-seam potency has decreased notably, but thanks to his cutter, his K/9 (9.42) and BB/9 (2.20) remain elite skills, and he's still delivering a top-shelf swinging-strike rate (13.9, significantly above the Fangraphs average of 8.9). But his mistakes are costly and, as noted in our previous in-depth look, are reaching outfielders more frequently.

The Rox look like they'll man a booth at the July 31 flea market and still have plenty of positives to sell regarding the righty. Betancourt was on one exec's list of potentially available relief arms, per ESPN.com's Jayson Stark.

Now for a non-Betancourt bullpen: Belisle isn't as dominant as Brothers, but his lack of walks and pattern of deployment in eighth-inning holds that protect save situations indicates he'd be the first choice. Brothers has more upside for fantasy help with his plethora of K's, though, so he'd probably factor in on some opps, if not take over eventually.

Joe Maddon confirmed what most of the fantasy world assumed: Fernando Rodney will remain closer even after the return of Kyle Farnsworth (elbow), which could come this weekend. Rodney's revelation is for real, and while he'll probably come back to earth a tad, he'll have plenty of rope.

Though it may take more than Thursday's blown save to erase Brett Myers' real-life trade value, it didn't help that it marked the second time in his last five outings he gave up at least five runs. His velocity has been OK, but he has fanned just five in 9 1/3 June frames.

This time, only one of six tallies was earned, but the four hits allowed shows how hittable he's been lately: 6.75 ERA, 2.04 WHIP. He plunked Carlos Quentin to force in a run and gave up a grand slam to pint-sized Alexi Amarista.

There's a palpable chance Myers won't be a closer after July 31, but if he continues to struggle, GM Jeff Luhnow might find it difficult to ditch him. Plan for both outcomes with your own squad and refresh yourself on the Houston Astros' speculative train. The front car is Brandon Lyon.

Rafael Soriano's job security was boosted Thursday, and he didn't even pitch. The New York Yankees gave him the night off after he pitched four of the last five games. Joe Girardi tried to piece together a save chance with Cody Eppley and Clay Rapada.

When they didn't work, he was forced to go to David Robertson, whom he's trying to preserve because of his recent return from the DL; the righty appeared on Wednesday, as well. After the two setup guys each put a runner on - one from a throwing error - D-Rob surrendered a three-run homer to lose the lead and the game.

Robertson is worth owning everywhere, but it doesn't look like Soriano, despite recent struggles, is in imminent danger.

Oil Futures Drop Again on Renewed Fears About Euro Zone

U.S. oil futures tumbled sharply Thursday on renewed fears that this week's euro-zone summit would again fall short of resolving the crisis.

Nymex oil futures settled at $77.69 a barrel, down $2.52, or 3.1%.

The fall came as European leaders converged on Brussels for another summit to try to resolve that region's sovereign debt crisis. Germany's Finance Minister signaled somewhat greater willingness than before to endorse debt mutualization. At the same time, yields on Spanish and Italian bonds have risen in recent days.

"The overriding issue is renewed liquidation related to equity weakening and the apprehension over the euro," said Jim Ritterbusch, president of trading advisory firm Ritterbusch & Associates.

"While it remains possible that the European leadership will pull a rabbit out of their collective hat, market expectations remain low," Credit Suisse said in a note.

Oil prices typically track economic expectations because of the commodity's central role in industrial production. Oil prices have retreated some 25% in recent weeks as hopes have dimmed about a speedy economic recovery.

Oil prices were trading lower throughout Thursday's session, but they nose-dived especially after a long-awaited U.S. Supreme Court ruling that largely upheld President Barack Obama's health-care law. Analysts said the relationship between oil prices and the health care ruling was indirect in that oil prices retreated in response to the fall in equity markets after the ruling came down.

The S&P 500 Index was trading at 1315, down 1.3% at 2:50 pm EDT, after hitting an intraday low of 1313 earlier.

Analysts said the markets remain jittery over demand weakness. A report by the Federal Reserve Bank of Kansas City Thursday gave fresh evidence of manufacturing woes.

The Kansas City Fed's manufacturing composite index--an average of the indexes covering production, new orders, employment, delivery times and raw-materials inventories--fell to 3 in June from 9 in May.

"Concern over demand issues continues to overshadow these markets," said Andy Lebow, a commodities broker with Jefferies Bache. "The market may still have some downside here."

While also trading lower Thursday, WTI's sister commodity, Brent Futures, was outperforming the U.S. benchmark somewhat. Brent prices were at $91.67 Thursday, down $1.83. The Brent market has picked up some strength amid a strike of Norwegian oil workers that has so far taken about 250,000 barrels a day off line. A note by J.P. Morgan observed that the Norwegian government was unlikely to intervene in the strike. The strike could "act as a support for Brent market structure," J.P. Morgan added.

Front-month reformulated gasoline blendstock, or RBOB, settled at $2.614 a gallon, down 0.24%. Heating oil settled at $2.55 a gallon, down 1.6%.

In this post I will discuss the role of human Hv1 channels in promoting the pathology and invasiveness of breast cancers. It is well known that cancer consists of normal cells gone awry. As cells age, they can accumulate mutations in their genomes and sometimes these mutations lead to unregulated growth and expansion. The unregulated, cancerous cells require a great deal of energy to produce their expansive growth and therefore many cancers are characterized by increased levels of metabolism, specifically of glycolysis. Because cancerous cells become dense tumors, the microenvironment inhabited by the cells can be very low in oxygen (hypoxic). High metabolism plus hypoxia means that the cells quickly start to build up high levels of protons (a byproduct of anaerobic glycolysis) in their cytoplasm (Gatenby & Gillies, 2007). Without a mechanism for dumping protons out of the cytoplasm, cancer cells would not be able to efficiently proliferate. When a cancer cell is able to efficiently proliferate it can spread from its primary site to other locations throughout the body, a process known as metastasis (for more information on this process check out metastasis on wikipedia). People are only beginning to tease apart the processes employed by different cancers to regulate their internal pH and become highly metastatic. One mechanism that is becoming apparent in certain types of breast cancer is the over-expression of human Hv1. Here I will discuss two recent papers by?Wang?et al.?that try to dissect the role of human Hv1?in breast cancer metastasis and clinicopathology (Wang?et al., 2011;?Wang?et al. 2012).

Human Hv1 is overexpressed in highly metastatic breast cancer cell lines

The discovery by Wang?et al.?of specific expression of human Hv1?in highly metastatic breast cancer cells?both in human tissue samples obtained from breast cancer surgery and in breast cancer derived cell lines?has led to the hypothesis that these channels are responsible for the regulation of cytoplasmic pH in these cells?(Wang?et al., 2011). What is most interesting from this first study is the specificity of the expression pattern, in which high levels of human Hv1 expression is only seen in the highly metastatic breast cancers and cell lines (Wang?et al., 2011). This suggests that the presence of the channel in the membranes of cancer cells may help them be more invasive.

To further investigate this possibility, Wang?et al.?generated a series of short interfering RNAs (siRNAs) that they used to reduced the expression of the channel in the cancer cell lines?(Wang?et al., 2011). When a highly metastatic cell line was treated with one of these siRNAs, it showed a significant decrease in migration and invasion, two hallmarks of highly metastatic cells. They also showed that the internal pH of the cells decreased, indicating an inability of the cells to properly remove excess protons from their cytoplasm (Wang?et al., 2011).

It has been demonstrated that the activity of the gelatinase proteases in the extracellular matrix (ECM) both facilitates the metastasis of cancer cells and is pH-regulated (Giannelli & Antonaci, 2002; Gioia et al., 2010). It is thought that these enzymes remodel the ECM, which in turn promotes growth and metastasis of the tumor. Wang?et al.?also show, using their human Hv1 siRNAs, that the activity and expression of these enzymes are reduced when the expression of human Hv1 is knocked down.

Although this initial study suggests that the presence of human?Hv1 is correlated to metastasis?and even suggests a mechanism through which disrupting pH regulation reduces migration and invasion; and disrupts the activity of gelatinases?Wang?et al.?are missing some very important control experiments. What they are missing is a ?rescue? experiment, i.e., they do not demonstrate that, by reintroducing an siRNA-resistant human Hv1?into the siRNA-treated cells, the migration, invasion and gelatinase activity levels return to control levels. In addition, they do not show whether, by introducing human Hv1?to a poorly metastatic breast cancer cell line, they increase these cells? ability to migrate and invade surrounding tissues. Without these controls we can?t be sure whether the effect shown in this study is directly from the activity of the Hv?channels or from some secondary, off-target effect of the siRNA. Without these rescue experiments, Wang?s study remains inconclusive.

Overexpression of Hv1 correlates with breast cancer clinicopathology

In a follow up study, Wang?et al.?examined 105 different breast cancer tissues taken from patients who underwent routine curative surgery (Wang et al. 2012). Each tissue sample was paired with a sample of normal breast tissue from the same patient and both samples were examined for overexpression of human Hv1?channels in a blinded manner (i.e. the clinical pathologist examining the sample did not know any of the clinical information prior to examination)?(Wang?et al. 2012). This large data set contained several different types of breast cancers and showed a wide range of human Hv1?expression. What was determined was that high levels of human Hv1 correlated with poor prognosis. In other words, the more Hv channels expressed in the tumor cells, the more deadly the cancer (Wang?et al. 2012).

Some of the other data in this paper is very similar to that discussed above for their 2011 siRNA study (Wang?et al., 2011). However, here they probe the physiological role that Hv?channels may be playing in the cells in more detail. Using?pH sensitive?fluorescent dyes, they show, that when human Hv1?is knocked down the ability of the cancer cells to regulate their cytoplasmic pH is decreased. In addition, the cells? ability to acidify the extracellular medium also decreases (Wang?et al., 2011). Again, they show that cells treated with Hv1 siRNAs have reduced gelatinase activity (Wang?et al., 2011), and again the control rescue experiments were not performed.

In the final experiment of the study, Wang?et al.?perform xenografts of the cultured tumor cells onto immunodeficient ?nude? laboratory mice (check them out on wikipedia). They show that the tumors that grow on the mice are smaller if the cells are pre-treated with the human Hv1 siRNA. They also show that the?tumors derived from the cells treated with?siRNA continue to have low Hv?channel expression levels?(Wang?et al., 2011).

Conclusions

These studies clearly show that human Hv1?is overexpressed in certain breast cancers and that the level of?Hv channel expression is correlated with prognosis. The more?Hv?channels a tumor has, the more likely that tumor is to be highly metastatic and deadly. However, a causal link between Hv?channels and breast cancer severity is not established. The experiments using?siRNA knockdown of human Hv1 are inconclusive. There are well-established standard for these sorts of RNA interference (RNAi) knockdown experiments and you must include proper controls to prevent the occurrence of false positives (Echeverri et al., 2006). Wang et al. fail to perform these controls and hence do not conclusively show that it is in fact Hv?channels that are responsible for the increase in cancer severity.

Off-target effects of siRNAs are more common than people are generally aware of and have led to the re-evaluation of high-profile papers several times (Hubner et al., 2010; Westhorpe et al., 2010). Regardless of this, every proper experiment needs its proper control. In siRNA studies, using a ?scrambled? or irrelevant siRNA (e.g. GAPDH, lamin B, GFP) is not enough: rescue experiments should be the minimal standard.

It is clear that in order to be highly proliferative cancer cells need to develop mechanisms to maintain their cytoplasmic pH. It is reasonable to assume that overexpression of Hv?channels would help provide cells with such a mechanism. This reasonableness and the correlation seen between Hv1?overexpression and clinicopathology certainly make an intriguing argument for the direct role of the channel in promoting metastasis. But as scientists we have to be skeptical. Before spending millions of dollars to develop specific?Hv?blockers for cancer therapy, I would certainly like to see the control experiments. However, if the control experiments are performed and demonstrate that Hv?current directly contributes to metastasis, then drugs that target and block?Hv?channels could be very promising in the future treatment of breast cancer.

Drake/Chris Brown Brawl Club Loses Liquor License

New York State Liquor Authority charges W.i.P. with 17 violations, saying the club poses a 'threat to public health and safety.'

The fall-out from June 14's bottle-bashing brawl reportedly between Drake's and Chris Brown's camps continues.

On Tuesday, The New York State Liquor Authority ordered an emergency suspension of W.i.P.'s liquor license, the club where the incident took place. W.i.P., with its adjacent (and affiliated) club Greenhouse, were both slapped with 17 separate violations.

The New York Daily News is reporting that R. Kelly, New York Knicks basketball star Iman Shumpert and the cast of the upcoming film "Savages" all had scheduled parties at the clubs, but now will have to look for new venues.

"The SLA will not tolerate violent bars that break the law," SLA Chairman Dennis Rosen said in a press release obtained by MTV News. "This should serve as a message that this agency will not hesitate to take immediate action when licensees pose a threat to public health and safety."

The release cites 10 separate assaults and altercations at the two clubs, the earliest occurring on March 11 and the latest being the bottle-throwing incident which left quite a few patrons injured, including NBA star Tony Parker, who is now suing the club for $20 million after he suffered a cut in his left eye.

While both Drake and Chris Brown and their respective entourages have been implicated in the brawl, police have yet to charge any of the men with a crime. Meek Mill, whose has also seen his name circulate in the brawl rumors, will appear on Wednesday's "RapFix Live" and address the drama starting at 4 p.m.

Communication: The #1 key to relationship management

Dr. Alan Zimmerman?s Personal Commentary:

Immediately after World War II, the Allied Armies gathered up many hungry, homeless children and placed them in large camps.? There, the children were abundantly fed and cared for.? However, at night the children did not sleep well.? They seemed restless and afraid.

Then one psychologist hit on a solution.? After the children were put to bed, each one of them received a slice of bread to hold.? If they wanted more to eat, they were given more food.? But this slice of bread was to be held and not eaten.

The slice of bread produced marvelous results.? The children would go to sleep, subconsciously knowing and feeling they would have something to eat the next day.? That assurance gave the children the assurance they needed so they could sleep calmly and peacefully.

In a similar sense, we all need something to hold on to.? And few things are more important or more helpful than holding on to good, healthy, positive, productive relationships at work or at home.

The good news is ? you can strengthen or manage almost any relationship ? to make it better and better.? The key to it all is the ?excitement principle.?

As I discussed in last week?s ?Tuesday Tip,? the research makes it clear ? anything that increases the excitement level of two people ? and is seen as a positive experience by both people ? will strengthen the relationship between the two of them.? In other words, whatever increases your heart rate, blood pressure, and adrenalin ? in a good way ? when you?re with another person or a team of people ? will build up your relationship.

I gave you five ways to do that last week.? However, the very best way to use the ?excitement principle? to strengthen and manage your relationships is to increase the quality of your communication.??

And most day-to-day talk in work and home relationships does not strengthen the relationship.? It is merely ?functional? talk ? such as ? ?Customer Y needs that product shipped today ? We?ve got until this afternoon to finish the project ? The kids will be home at 5 ? and ? Let?s have pizza for dinner.?? It gets a job done, but it does not increase the excitement level or bring the people any closer together.

So what kind of communication works?? There are four kinds, each one more powerful than the previous one.

1.? Talk about the past.

Talk to the other person about things you did together in years gone by.? Talk about the places you?ve been together.? And talk about the things you got excited about.

This works equally well for teams on the job or family members at home.? When you talk about ?the good old days? or ?how things used to be,? you rekindle a certain camaraderie.? You see it happening at the annual company picnic or the ten-year class reunion.? It just plain feels good to remember all the good and bad things you went through ? together.

As you do so, remember to follow English author Quentin Crisp?s advice.? He said, ?The formula for a successful relationship is simple:? Treat all disasters as if they were trivialities, but never treat a triviality as if it were a disaster.?? In other words, put things in the best possible perspective.

2.? Laugh together.

Laughter releases endorphins that kill off pain.? And these endorphins can lead to a sense of well-being and optimism..? You benefit personally.

But when you laugh together, you also strengthen and manage your relationship more effectively.? The laughter can bridge the gap between total strangers and laughter can even reduce the tension in a tense situation ? which is often needed ? on and off the job.?

So I?m always looking for humor I can share with my friends and family as well as my clients and audiences.? For example, one of my colleagues, Scott Friedman, sent me the following.

  • A chicken crossing the road is poultry in motion.
  • Shotgun wedding: a case of wife or death.
  • Reading while sunbathing makes you well-red.
  • When two egotists meet?it?s an I for and I.
  • What?s the definition of a will? It?s a dead giveaway!
  • If you don?t pay your exorcist, you get repossessed.
  • Once you have seen one shopping center, you?ve seen a mall.
  • The man who fell into the upholstery machine is fully recovered.
  • Local Area Network in Australia: The LAN down under.
  • Those that get too big for their britches will be exposed in the end.

As fun and helpful as that can be, it?s even more effective if you ?

3.? Engage in open, honest self-disclosure.

Reveal some things about yourself that the other person would not know otherwise ? things that involve a bit of risk.? It kicks the ?excitement factor? into play.? After all, there is no intimacy without transparency.? And there is no teamwork without self-disclosure.

And one of the best places for self-disclosure to take place is in the performance appraisal process.? Unfortunately, many organizations have a difficult time getting their managers to give their employees timely, helpful performance appraisals ? because the appraisals provoke too much anxiety.?

But that?s crazy.? If a manager has been doing his/her job ? self-disclosing and sharing information with the employees on a regular basis ? there shouldn?t be any anxiety in giving performance appraisals.? All the appraisal does is summarize all the information the manager has already given the employee about his/her performance.

The anxiety comes into play when the manager stores up information for long periods of time.? Again, that?s crazy.? In so doing, he/she takes away the employee?s chances of improving his/her performance.? And what right does any manager have to deny his/her employees the opportunity to do the best possible job?? A manager has to self-disclose and give complete and honest feedback.

As a rule of thumb, if you?re a supervisor or manager, give your subordinates feedback every month that tells them how they?re doing.? Better yet, do it every week.? And when you have that brief discussion, ask your subordinates for some feedback.? Ask them if there is anything you could be doing more effectively.? Ask them if there is anything you could be doing to help them with their jobs.? You will be managing the relationship in way that allows for more back-and-forth self-disclosure.

Of course, there is this myth that feedback should only flow downward in an organization.? Actually, the best ? and maybe the most important ? feedback flows upward.? The Nixon White House needed to learn that.? Nixon created an environment where his subordinates only told him what he wanted to hear.? They did not give him any opinions that were contrary to the views he already held.? Of course, it created insanity.?

And there is a measure of insanity in any organization when the upper-level bosses are deprived of feedback.? They lose touch with reality and become very much like the Emperor and his new clothes.? No one told him the truth; so he walked around naked.

In my speaking and consulting practice, I find that the worst managed companies are those where it is common knowledge that it is NOT okay to go into the offices of a senior executive and give him/her absolutely honest feedback.? Doing so might result in the loss of the employee?s job or at least damage his career.?

I tell executives that it is YOUR job to create an environment of trust that ripples throughout every level of the organization.? It is YOUR job to create an environment where subordinates feel safe giving you open, honest feedback.?

The fact is ? everyone benefits when leaders, managers, and subordinates are open, honest, and self-disclosing.? In fact, I?ve noticed that is the norm in the best organizations.?

And how do you know if you have that kind of an environment going on at work?? Simple.? On a piece of paper, write down the name of your superior.? List all the things he or she does well and all the areas where improvement is needed.? Then imagine yourself bringing that paper to your superior.? If you feel any anxiety about that, it?s because there are some things on your paper you haven?t told your superior.

In essence, to strengthen and manage your relationships, increase your self-disclosure.

And the VERY BEST way to do that is ?

4.? Ask and answer brave questions.

It?s a skill that is lacking in so many relationships.? Lots of people don?t know how to connect on a deeper level.? So they talk about the things they have to do, the people they know, or relatively unimportant topics such as the weather, the football game, and what?s for dinner.?

But the strongest teams and best relationships always go beyond superficial conversation.? They ask what I call ?Brave Questions? ? questions that are a bit more personal ? and a bit more risky.? They seek to understand the other person on a much deeper level.? And when they?re done right, you infuse your relationships with a surge of energy.? You go beyond relationship management to relationship building.

Deb Olswold testified to that.? She wrote to tell me this technique got her a better job.? In her words, Deb said, ?I have always been a very shy person, always in the background of the workplace, the family, and my personal life.? Then I was asked to interview for a higher ranking position, so I rushed out to get a copy of your book, ?Brave Questions.?? I read the book, wrote a few of the questions on a small piece of paper, and went to the interview. Instead of just passively sitting back and letting them take the entire lead, I asked them such things as, ?Where do they want to be in two years?? What kind of person are they looking for?? And what do they expect from me???

?Because of your book, I went through the entire interview with a sense of confidence.? An hour after the interview, they called to offer me the position.? I accepted the position and have thanked you in my daily prayers.? I know that your ?Brave Questions? helped me get the best job I have ever had. The 400 questions in the book are fantastic.? Action:

Decide this week to engage at least two people in a deeper conversation where you ask each one of them at least two Brave Questions. And then listen closely to their answers.

The role: Manager, SMB Sales, Central and Eastern Europe (CEE)

With a rapidly growing array of sales teams, it's important to have seasoned, charismatic leaders who guide our client sales teams to success. As a Sales manager, you lead and coach high-performing account teams that use consultative sales skills to understand our advertisers' needs and deliver measurable solutions. You also work with the Sales leadership team to set strategic objectives and run the day-to-day operations for the business. Sales managers help their teams deliver on ambitious targets, and roll up their sleeves to partner directly with clients to exceed revenue and merchant growth targets across markets and verticals. You are an excellent communicator with a proven ability to train and motivate a large team, and you take an analytical approach to sales management.

Responsibilities:

  • Represent the SMB Sales Team in target setting for your region and market. Take ownership for delivery of revenue target for your market and sales program devising strategies to meet quarterly quotas.
  • Develop and implement best practices for your sales programs and markets. Ensuring that your team is always up to date with the newest advertiser solutions.
  • Work with and own projects among the wider management team to deliver cohesive and co-ordinated strategy development of the sales programs.
  • Manage a team of SMB sales representatives and team leads, working on the growth of Google's advertiser. Coach and manage your team to develop sales and account management skills and contribute to the management of the wider market team.
  • Execute projects involving quantitative analysis, industry research and strategy development.

Minimum qualifications:

  • BA/BS degree (In lieu of degree, relevant skills or equivalent experience).

Preferred qualifications:

  • Extensive sales management experience in an online business or targeting the CEE market, in particular Russia. Proven ability to leverage account management, relationship building and planning to increase revenue from existing sources.
  • Experience leading teams, with a strong focus on coaching and mentoring, and demonstrated success developing sales teams and hitting revenue targets.
  • Proven ability to manage multiple projects while paying strict attention to detail. Self-starter who is comfortable with a rapidly-changing environment and working to targets.
  • Ability to think strategically, and ability to make changes to meet strategic needs within your market.
  • Strong communication skills in English as well as a CEE language, in particular Russian.
